BL21 is the most often used host strain for protein expression with the T7 polymerase / prophage DE3 system of Studier and Moffatt (1985). Unfortunately the means and the results of the genetic transduction by which the B834 strain became Met+ are not documented by Studier and Moffatt. Due to this lack of defined pedigree most of the BL21 based expression hosts are not the best choice when it comes to preparing the documents for approval by an pharmaceutical authority.
